If powering by USB, the +5V is sourced from the USB and +5 is regulated to 3.3V output. BUT, the Barrel connector is directly connected to +5V output and this value regulated to 3.3V output. So, if connecting, for example, +12V to barrel connector, then +5V output will actually be +12V. Also, barrel connector is connected directly to USB so don't plug in USB at same time as barrel.
